PERFORMANCE DATA SHEET 0840 Monoclonal anti-human CD122 (IL-2Rb)/Biotin*
mAb name/Clone:9A2 (DU-2) Isotype: Immunogen: YT-1 acute human leukemia cells
CATALOG#: 343-030 QUANTITY: 100 mg
CONCENTRATION: 1.0 mg/ml INFORMATION: Human CD122 (IL-2 Rb) associates with CD25 (IL-2Ra) to form a high affinity receptor for IL-2 and is expressed on activated T cells, B cells and monocytes. The cytoplasmic domain of CD122 is involved with signal transduction. Antibody
9A2 is specific for the 75 kd b subunit CD122 of the high affinity interleukin-2 receptor. Antibody 9A2 (DU-2) inhibits binding of IL-2 to IL-2Rb
(CD122). References: STORAGE CONDITIONS: Store at 2 - 5oC. Freeze/thawing not recommended. PRODUCT STABILITY: Product should retain activity for at least 6 months after shipping date when stored as recommended. Ship Date:_____________ BUFFER: 50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, 5% Glycerol, 0.2% BSA, 0.04% NaN3 (as a preservative). PRODUCTION: Antibody from (low FBS containing) tissue culture supernatant was Protein A purified to >95% mouse immunoglobulin by SDS-PAGE (<1% bovine immunoglobulin), and reacted with NHS-Biotin. Unconjugated Biotin was removed from conjugate using a desalting column. PERFORMANCE: Briefly, 5 x 105 ficoll prepared human peripheral blood lymphocytes were stimulated by incubating 4 hours with 5 mg/ml PHA-P(Sigma). Stimulated cells were harvested by gentle scraping, washed and incubated 45 minutes on ice with 80 ml of anti-CD122/Biotin at 20 mg/ml. Cells were washed twice and incubated with 2o reagent Streptavidin R-Phycoerythrin (Catalog #253-050), after which they were washed three times, fixed and analyzed using a BD FACstar plus. A net 17.1% population of the cells stained positive with a mean shift of 0.28 log10 fluorescent units when compared to a tube where cells were pre blocked 10 minutes with 20 ml of 0.5 mg/ml anti-CD122(IL-2Rb) antibody (Catalog #343-020).
